INTRODUCCIÓN
Numerosos métodos, entre otros los descritos por Dellaporta et al. (1983),
Guillemaut y Marechal (1992), Honeycutt et al (1992), Hoisington et al. (1994),
Harvey y Botha (1996), Aljanabi y Martínez (1997) y Chen y Ronald (1999) han
sido desarrollados para diferentes especies de plantas, sin embargo, la mayoría
requieren una alta inversión de tiempo, material vegetal y reactivos, o producen
bajo rendimiento de ADN.
Cuando se trabaja con un gran número de plantas como es el caso de los bancos de
germoplasma se requiere un método rápido y eficiente de extracción que produzcan
ADN de alta calidad.
MATERIALES Y MÉTODOS
Material vegetal
Se utilizó tejido foliar joven de Saccharum spp., Musa sp., y Manihot esculenta, al
cual se le realizó el procedimiento modificado de extracción de ADN descrito por el
CIAT (1999) para Pyricularia grisea en arroz.
7. Lavar el pellet de ADN dos veces con 500 μl de Etanol al 70% frío. Descartar el
Etanol y permitir que el ADN se seque al aire libre por 30 min.

SUMMARY
Numerous method for extraction of DNA have been developed for different
plant species, however, the majority require a high investment of time,
plant material and reagents, or produce low yields of DNA. When working
with a great number of plants as with germoplasm collections, a fast and
efficient method of extraction to produce DNA of high quality is required.
The methodology describes for DNA extraction is fast, simple and renders
genomic DNA of good quality, free of secondary compounds, proteins and
polyphenols that coprecipitate with the DNA inhibiting the reaction of DNA
amplification. Methodology produced between 15-25 μg per 100 mg of
fresh tissue. Quality of DNA was validated through its consistent
amplification using RAPD technique in Saccharum spp., Musa sp.,
and Manihot esculenta. Time required for DNA extraction was reduced to a
40% and its cost and level of toxicity to 50%, compared with other
commonly used methods.
