Las rutas deWnt, Shh y Notch contribuyen a la autorrenovación de células troncales en varios órganos,así como en el SN y en el
hematopoyético. La desregulación de estas rutas puede contribuir a la oncogénesis. Se han descrito mutaciones de estas rutas en varios
tumores humanos: carcinoma de colon y tumores epidérmicos (Wnt), meduloblastoma y carcinoma de célulasbasales(Shh) y leucemias
de células T (Notch).
