En una transición de primer orden se observa que Cp = ( H / T)P es diferente en las dos fases. Cp
puede aumentar (como en la transición de hielo a agua) o disminuir (como en la transición de

Se usan rayos-X de longitudes de onda conocidas.
Generador de rayos-X (ANODO ROTATORIO)
Colecta de datos de difracción de rayos-X. Actualmente se
usan crio-protectores para proteger a los cristales del daño por
radiación. La colecta se hace a T del N2 liquido (~ -160 °C)
PATRON DE DIFRACCIÓN DE UNA PROTEÍNA (los puntos muestran
las intensidades de difracción de los motivos estructurales (átomos
o moléculas) ubicados en planos cristalográficos).
Si los cristales son grandes con dimensiones de 100 micrómetros se pueden
colectar los datos en un equipo de rayos-X casero (ánodo rotatorio). Si son
menores a 50 micrómetros se tiene que usar radiación sincrotrón. En los
sincrotrones los rayos-X son mas luminosos (intensos) y se producen en
instalaciones de aceleradores de partículas (electrones) por altas energías.

Purification of grenache berry PPO. (A) Gel filtration chromatography of combined ammonium sulfate pellets after dialysis was
applied to a Sephadex G-75 (167 × 1.7 cm2) column, equilibrated with 5 mM sodium phosphate, pH 8.0, and eluted with the
same buffer at a flow rate of 0.3 mL/min; 4.0 mL fractions were collected. The fractions containing peak 2 were combined for ion
exchange chromatography. (B) Anion-exchange chromatography using an Econo-Pac High Q cartridge (1 × 5 cm2). Elution was
obtained using a NaCl gradient from 0 to 0.2 M NaCl in 5 mM sodium phosphate, pH 8.0, for 136 min at a flow rate of 1.0 mL/
min, collecting 2.0 mL fractions. After 166 min, the gradient was increased to 1.0 M NaCl, eluting a second, smaller peak with
PPO activity. (C) HPLC chromatography of the fractions corresponding to peak 1 of part B on a Superose 12 HR10/30 column,
equilibrated with 10 mM Tris-HCl, pH 7.5. (D) SDS-PAGE. Lanes 1−4 contained fractions 1−4 from the HPLC experiment shown
in part C; lane 5, molecular mass markers. Upper gel: silver stain, fully denatured protein; lower gel: activity stain using 25 mM
4-tert-butylcatechol and 25 mM 4-amino-N,N-diethylaniline sulfate. Incompletely denatured protein was used for the activity gel,
and bands might not represent different molecular weights.
