La esferocitosis hereditaria es una enfermedad genética que afecta a proteínas de la
membrana del eritrocito, lo que provoca defectos en el citoesqueleto. La incidencia de
esta afección es de 1:5000. El defecto más frecuente es la deficiencia de espectrina,
que hace que algunas regiones de la membrana se rompan porque ya no están
ancladas al citoesqueleto. La membrana restante puede cerrarse de nuevo de forma
espontánea, pero después de varias «roturas» la célula se vuelve finalmente pequeña
y esférica. La presencia de estos microesferocitos es la característica distintiva de la
esferocitosis hereditaria, afección que puede ser asintomática o llegar a manifestarse
por una anemia hemolítica grave. La hemólisis (destrucción anormal de los eritrocitos)
se produce porque los esferocitos son especialmente sensibles al estrés osmótico.
