ABSTRACT: The general objective of this laboratory practice is to identify some amino acids and
their characteristic groups in the side chains by means of reactions that allow differentiating color
changes. In addition, the thin layer chromatography (TLC) technique is used to identify and separate
them due to their different affinities for the mobile and stationary phases. As a result of the different
reactions, different physical changes in the amino acids must be presented for their respective
identification, these processes are called qualitative reactions where, for example, reaction with
ninhydrin is used, among others. To separate and identify the amino acids, fresh spinach was used
and by means of the chromatography technique, using cellulose acetate as the stationary phase and
butanol-acetone-acetic acid as the mobile phase. In conclusion, there are several ways to identify
amino acids and also to separate them when dealing with a sample with a large amount of AA.

INTRODUCCIÓN:
METODOLOGÍA:

Se traza una línea de con ayuda de un lápiz en la fase estacionaria con una
ubicación de 1,5 cm del borde inferior. Línea la cual es denominada la línea de
siembra. Con ayuda de un tubo capilar se adicionan 5 gotas de extracto etanólico de
espinaca sobre las líneas de siembra junto con 3 gotas de cada aminoácido patrón.
Secar las gotas aplicadas en la placa con ayuda de una estufa y colocar
rápidamente la placa en la cámara de cromatografía verificando que los solventes
queden por debajo de la línea de aplicación y tapar.
Para finalizar se identifican todas las manchas con ayuda de un lápiz indicando el
centro de cada una de ellas para poder generar los datos que contribuyen para
calcular el Rf. Marcar, envolver en película plástica y llevar a destino final.
