In the granules obtained, three thermal events were detected by DSC: the fusion
of PEG, the dehydration of lactose and the fusion of the drug; It was also
observed that by adding crospovidone to the mixture, the fluidity of the granules
increased, indicating a positive effect on the rheological properties. The
dissolution rate of the drug showed an improvement in vitro in all the granules
that were prepared, from 3.5 to 3.7 times. The different amounts of PEG used
showed no significant difference between each other in the dissolution rate. With
the granules that showed flow properties favorable tablets were compressed,
lubricating them with magnesium stearate at 141 2%. In all cases good dough
uniformity was shown. Tablets without intragranular crospovidone did not comply
with the disintegration times established in the European Pharmacopoeia;
However, this was solved by adding the crospovidone extragranularly.
Conclusion The low solubility of drugs conditions their absorption. Most of the new chemical
entities are poorly soluble in aqueous solvents, which causes the low
bioavailability of these drugs and therefore obstruction of the therapeutic effect. It
is possible to enhance the biopharmaceutical characteristics of drugs if adds more
than one polymer and/or a surfactant in the formula. In the formulation of SD, it
is important to correctly select polymers that meet the appropriate conditions. It
is suggested to consider hydrophilic polymeric carriers that are amorphous and
that have low Tg values, which allow the dispersion of the drug in its amorphous
form and that form a monophasic dispersion.
The literature review shows that SDs manufactured in HSM are a successful
method to ensure that BCS class II drugs modify their physicochemical properties
solubility and dissolution rate. Regarding the manufacturing process, there is a
wide variety of methodologies for manufacturing DS. HSMs have been
successfully used to manufacture DS by various methods, such as granulation via
wet, solvent evaporation and melting. The precedent in the use of HSM in the
pharmaceutical industry for conventional granulation is an advantage, because it
eliminates the infrastructure investment costs derived from the purchase of more
sophisticated equipment, such as hot melt extruder or spray drying bed.
