PRESIÓN HIDROSTÁTICA

• Según la ADA se considera alimento funcional debido a su fuente de
carotenoides, vitaminas y personas. Además reduce el colesterol LDL,
mejora salud cardiovascular, previene el cáncer y regula niveles de
glucemia en la sangre.

OBJETIVOS

 Evaluar la estabilidad de los carotenoides de la pasta de aguacate, en el proceso

HHP y durante su almacenamiento.
 Evaluar la ORAC ( Capacidad de absorbancia de radicales de oxigeno hidrófilos y
lipófilos) durante HHP y el almacenamiento.
 Comprender la estabilidad de diferentes moléculas bioactivas propias del aguacate.
MATERIALES Y METODOS
Para la elaboración del análisis se adquirió Luteína, β-ciclodextrina, KOH, NaCl, acetona, hexano,
metil butil éter, agua , metanol e isopropanol.

1. PREPARACIÓN DE LA MUESTRA Y HHP

- Lavado, desinfección, pelado y macerado. Se desairó (-88,2 kPa), se empaco al vacío y
se llevo a 5 C.
2. Se hizo HHP a 600 Mpa durante 3,5 minutos y 2,75 minutos duro la descompresión.
3. Se llevo a almacenamiento a 4 C.

Mediciones:
LOX= Se mide por formación de dienos, con acido linoleico se extrae la enzima y se mide
espectrofotometría a 234 nm.
Oxigeno disuelto= Se mide por electrodo.

4. IDENTIFICACIÓN Y CUANTIFICACIÓN DE CAROTENOIDES

Extracción con BHT, en oscuridad, homogenización con acetona, filtrado (tres veces),
concentración del extracto (Evaporador centrifugo), saponificación (KOH), dilución con agua,
decantación con hexano, suspensión con isopropanol y separación con membrana.

4.1 Análisis por HPLC-PDA

4.2 Preparación de Curvas Estándar
Trans-β-caroteno, β-apo 8'carotenal y luteína.
Soluciones madre en hexano y etanol.
Se realizo espectrofotometría (a diferentes longitudes).
Se separara con evaporador centrifugo y suspendió con isopropanol
Separación con membrana.

4.2 INTERPRETACION DE ESPECTROS UV-VISIBLES

Identificar el espectro y comparar con informes previos.
Identificacion con tiempo de retención y características con normas comerciales.
Identificar por orden de elución reportado en la literatura, a las mismas condiciones
cromatografía.

5. DETERMINACION DE L-ORAC Y H-ORAC

Liofilización, homogenización con hexano, baño de ultrasonido, filtrado al vacio (extracto de
hexano). Se pone en baño de agua (evaporar hexano) y se disolvió con acetona.
El filtrado se homogenizo con AWA, baño ultrasónico y filtrado al vacio (3 veces).

Determinacion de L-ORAC y H-ORCA con lector de microplacas a 37 C.

RESULTADOS
Disminución en los niveles LOX ~44%, literatura (~43-55%).
Niveles de oxigeno de ~15 ppm.

Aumentos de carotenoides
individuales, luego de la presurización CAROTENOIDE AUMENTO
Neoxantina-b 513%
Criptoxantina α 312%
Criptoxantina β 220%

CONCLUSIONES

 Los carotenoides aumentaron al final de la vida de anaquel sensorial del

producto en comparación a la pasta sin procesar mientras disminuyeron en el
almacenamiento.
 Evaluar la ORAC ( Capacidad de absorbancia de radicales de oxigeno
hidrófilos y lipófilos) durante HHP y el almacenamiento.
 Comprender la estabilidad de diferentes moléculas bioactivas propias del
aguacate.
