De aqui en adelante en esta descripcién, el término “particulas finas” incluye polvo pulverizado (polvo en particulas finas). ANTECEDENTES DE LA TECNICA En general, con respecto a la administracién transpulmonar, se sabe que el ingrediente activo contenido en una medicina puede administrarse a los Pulmones de manera eficiente haciendo que el diémetro de las particulas del ingrediente activo sea 10 micrémetros © menos, de preferencia 5 micrémetros o menos. Asi, la situacién actual con inhalaciones convencionales para administracién transpulmonar es que, para hacer que la medicina tenga un didmetro de particulas adecuado para la administracién transpulmonar por adelantado, sePagina preparan particulas finas por el método de secado por atomizacién, un método de trituracién de chorro o similer, y posiblemente se lleva a cabo un mayor Procesamiento, y luego las particulas finas se proporcionan en un inhalador de polvo seco. Especificamente, la Publicacién de Patente Japonesa No Examinada No. 1999-171760 divulga tres tipos de inhalacién en polvo, a saber, (1) una preparacién que comprende una composicién en forma de polvo que comprende s6lo particulas finas medicinales dentro de un vehiculo adecuado, (2) una preparacién que comprende una composicién en forma de polvo en la cual se han granulado particulas finas medicinales ligeramente para formar un diémetro de particulas relativamente grandes contenidas en un recipiente adecuado, y (3) una preparacién que comprende una composicién en forma de polvo que comprende Pparticulas mezcladas en la cual particulas finas y particulas de recipiente (lactosa, etc.), que tienen un diametro mayor que las particulas finas medicinales se mezclan de manera uniforme dentro de un recipiente adecuado. © Adem’s, se ha divulgado que si estas imhalaciones en polvo se administran en el tracto respiratorio, el comportamiento que se muestra es que con (1) las particulas finas medicinales en la composicién % 7 5Fa Pagina oA llegan al tracto respiratorio inferior, por ejemplo, la tréquea y los bronquios, y se depositan en éstos, con (2) 1a medicina granulada se separa en particulas finas en vuelo en el tracto respiratorio, y Ias particulas finas medicinales producidas llegan al tracto respiratorio inferior, por ejemplo, la tréquea y los bronquios, y se depositan en éstos, y con (3) el vehiculo se deposita en la cavidad oral, en la faringe o en la laringe, y las particulas finas medicinales sé6lo llegan al tracto respiratorio inferior, por ejemplo, la trdéquea y los bronquios, y se depositan en éstos. De esta manera, con una inhalacién en polvo convencional para administracién transpulmonar, el ingrediente que va a ser inhalado se convierte en particulas finas adecuadas por adelantado, y luego estas particulas finas, o estas particulas finas mSs procesadas por algin otro método, se llenan en un inhalador de polvo seco, y la administracién transpulmonar se realiza usando este dispositivo. Para convertir un farmaco de bajo peso molecular en particulas finas, por lo general se usa un método de secado por atomizacién (por ejemplo, un método divulgado en la Publicacién de Patente Japonesa no Examinada No. 1999-171760), un método de trituracién a chorro (porPagina ejemplo, un método divulgado en la Publicacién de Patente Japonesa no Examinada No. 2001-151673) o similar. #1 método de trituracién a chorro comprende aplicar un impacto de aire que tiene un indice de flujo de aire de al menos 1 000 L/min y una velocidad de aire no menor que la velocidad sénica para un ffrmaco de bajo peso molecular que convierte el férmaco en particulas finas. No se conoce ningin método que convierta el férmaco en particulas finas con un bajo impacto de aire. En el caso de un farmaco de alto peso molecular como un péptido o proteina, por el otro lado, por ejemplo un método en el cual una solucién para atomizacién de un liguido concentrado medicinal que contiene aditivos, se somete a secado por atomizacién, convirtiendo de este modo el liquido concentrado en particulas finas que tienen un didmetro de 5 micrémetros o menos en un paso, y luego estas particulas finas se llenan en un inhalador de Polvo seco (método de secado por atomizacién: WO 95/31479), y un método en el cual un péptido o proteina se seca por congelamiento junto con aditivos, y luego la composicién secada por congelamiento se convierte en Particulas finas por trituracién de chorro 0 similar, y el inhalador de polvo seco se liena con estas particulas DsPégina finas conocidas (método de secado por congelamiento - trituracién de chorro: WO 91/16038). No obstante, las inhalaciones de —polvo convencionales para = administracién = transpulmonar Preparados por los métodos de secado por atomizacién o secado por congelamiento - trituracién de chorro no son necesariamente preparaciones ideales para farmacos de alto peso molecular como péptidos y proteinas en particular. Por ejemplo, como se muestra en la divulgacién en WO 95/31479 que alrededor del 25% de desactivacién de interferén ocurre durante el proceso de secado por atomizacién, se anticipa que si se usa el método de secado por atomizacién, luego las proteinas y similares se desactivarén en el proceso de fabricacién y la actividad del farmaco disminuiré de este modo. No conoce ningin método que convierta un farmaco de alto p. jo molecular en particulas finas mediante un bajo impacto de aire, al igual que un farmaco de bajo peso molecular. Adem4s, con el método de-secado por atomizacién y el método de secado por congelamiento-trituracién de chorro, se requiere una operacién en la cual el polvo fino Preparado se recolecte del aparato de secado por atomizacién o aparato de trituracién de chorro, y seSe m Pagina 4 4 subdivida y llene en recipientes. De esto modo, es inevitable que junto con esta operacién surjan problemas . como que la el rendimiento de la preparacién disminuya debido a la recoleccién o pérdida de llenado y, por consiguiente, el costo se eleva, y que la preparacién se contamine con impurezas. Adem4s, en general resulta dificil subdividir y llenar con polvo pequefias cantidades con precisién. si se usa el método de secado Por atomizacién o el método de secado por congelamiento- método de trituracién de chorro, para el cual es esencial dicha subdivisién y llenado de cantidades pequefias en forma de polvo, resulta necesario establecer un método de llenado con pequefias cantidades y exactitud de polvo. Detalles de un sistema, aparato y método para el llenado de polvo fino se divulgan en la Patente de los Estados Unidos No. 5,826, 633. DIVULGACIGN DE _LA_INVENCIGN Constituye un objetivo de la presente invencion resolver varios problemas de las inhalaciones convencionales de polvo antes mencionadas para la administracién transpulmonar. Especificamente, constituye un objetivo de la presente invencién Proporcionar un sistema de preparacién y sistema de administracién nuevo que permitan que una composiciénPagina y secada por congelamiento que se encuentre en recipientes se subdivida por adelantado en dosis inicas de ingrediente activo para convertir las particulas finas a un didmetro adecuado para administracién transpulmonar en el recipiente en el momento de su uso y luego usarse para administracién transpulmonar como se encuentri los inventores de la presente realizaron minuciosos estudios para lograr el objetivo antes sefialado, y como resultado descubrieron que si la _—sustancia farmacolégicamente activa se deposita en forma de liquido en recipientes subdivididos en cantidades necesarias y luego se secan por congelamiento, la composicién secada Por congelamiento en forma distinta de polvo preparada de este modo puede convertirse de manera inesperada en particulas finas por un impacto de aire relativamente bajo mientras atin se encuentra en el recipiente. Baséndose en este conocimiento, los inventores de la presente realizaron otros estudios, y como consecuencia de éstos descubrieron que usando una composicién secada por congelamiento, una sola dosis de lo que se encuentra en forma distinta de polvo en un recipiente, en combinacién con un dispositivo que comprende medios para introducir aire a una velocidad e indice de flujo prescrit en el recipiente para ser capaz de aplicar un wi “aPagina Sf impacto de aire prescrito a la composicién, y medios para descargar del recipiente la composicién en polvo que se ha convertido en particulas finas, luego la preparacién secada por congelamiento puede ser preparada, en forma de polvo de particulas finas adecuada para 1a administracion transpulmonar, f4cilmente por un usuario en el momento del uso (especificamente el momento de 1a inhalacién), y el polvo de particulas finas puede administrarse por inhalacién tal como se encuentra. Asimismo, se ha verificado que de acuerdo con este sistema de administracién transpulmonar, se pueden resolver todos los problemas previamente mencionados respecto de las inhalaciones convencionales de polvo para administracién transpulmonar. Es decir, de acuerdo con el sistema de administracién transpulmonar de 1a presente invencién antes mencionado, no es necesario recolectar 1a Preparacién farmacéutica en forma de polvo y luego lienarla en recipientes, sino que la preparacién se realiza llenando cada recipiente con liquide y luego se efectiia el secado por congelamiento, y luego el sistema de administracién transpulmonar puede usarse para administracién transpulmonar con una precisién sumamente alta y una produccién alta de 1a preparacién, y sin el
