Monday, May 15, 2017

Week 7 is the busiest week of my On-the-Job training so far. Everyday there were
analysis. On Monday, six (6) samples were delivered to the laboratory. They were immediately
measured for Biological Oxygen Demand. Per usual, samples that are only measured for BOD
are directly transferred to fill up the BOD bottle, where one millilitre (1mL) of each MnSO4 and
alkali iodide were added.

After the bottles were shaken, the precipitate were allowed to settle until the it covered
only one-third () of the solution then one millilitre (1 mL) of concentrated H2SO4 was added.
After shaking the bottles, it was titrated with Na2S2O3, until pale yellow then a starch indicator
will be added and the titration continued until the solution became colorless.

Serial dilution bacterial analysis were also usually done together with the measurement
of BOD whenever there were new delivered samples. The method is still the same as described
before. Where it will start with the transferring of eleven milliliters (11-mL) of the pure sample to
the bottle containing diluent solution then it was shaken and placed on its original place.

Ten milliliters (10-mL) of the pure sample was transferred to the sample bottle containing
the LTB (Lauryl Tryptose Broth; for the detection of coliforms in water). The eleven milliliters
(11-mL) of the diluted sample is transferred to another bottle containing diluent solution then it
was shaken and placed on its original place. Ten milliliters (10-mL) of the diluted sample was
transferred to the sample bottle containing the LTB. the process continues until all the bottle
containing diluent solutions were used up.

This day, seven (7) samples were delivered to the laboratory. These samples were
measured for dissolved oxygen. After the samples were prepared and seeded, one millilitre
(1mL) of each MnSO4 and alkali iodide were added. The procedure from here are just the same
with the measurement for BOD.

For the bacterial analysis, serial dilution was still the method that was used.

For wednesday, there were fifty (50) samples where measured for dissolved oxygen.
Polyseed was prepared by dissolving it in distilled water and was stirred for an hour before
adding it to the sample bottles containing 15-20mL of the pure samples. It was then diluted
using the prepared dilution water. The procedure from here are just the same with the
measurement for BOD.

Since there were some days that work cant be done, many samples were not yet tested
for ammonia and phosphates. That is why there were forty-nine (49) samples were tested for
ammonia and thirty-six (36) samples were tested for phosphates. The first thing we did is
prepare Erlenmeyer flasks (50-mL) for the samples to be tested for ammonia and labelled them
before transferring twenty-five millilitres (25-mL) of samples there. One millilitre (1-mL) of
phenol-alcohol reagent, sodium nitroprusside and an oxidizing agent. The samples were left for
an hour or so to wait for the reaction the complete before reading it in the UV-VIS
Spectrophotometer.
For the test for phosphates, Erlenmeyer flasks (250-mL) were prepared and labelled
then twenty-five millilitres (25-mL) of samples where transferred. Ascorbic acid, 4M H2SO4 and
oxidizing agent were added. It was immediately read in the UV-VIS Spectrophotometer because
unlike the test for ammonia the reaction is a lot quicker.

Concentration in Water
After the test for ammonia and phosphates, measurement for dissolved oxygen was
done. It wasnt a lot so it was done quite fast. There were only 30 samples to be measured.
