Abstract

Studying and understanding the sources of variation in early life


traits in farmed deer are fundamental for management and/or breeding
purposes. Data from a captive white-tailed deer population were analysed to
identify non-genetic and genetic factors affecting the birth weight (BW) of
fawns. The year, type of birth and sex were included in a fixed linear model to
examine their significance. All of the examined non-genetic factors had a
highly significant effect on BW (P<0001). The examined years showed
variation attributed to food availability affecting the gestational conditions of
does. Male fawns were 193 g heavier than female fawns at birth (P<0001),
and singleton births were associated with a higher BW (2970043 kg)
compared with twin (0261 g) and triplet (0642 g) fawning (P<0001). The
best-fitting animal model was selected by comparing reduced and complete
models. Based on the selected animal model, which included direct genetic
and common maternal effects, genetic components and parameters were
estimated. The direct heritability was found to be 0280126, and a small but
important contribution of common maternal environmental effects was
identified (c2=0150062). The results support the importance of certain
environmental factors affecting BW and indicate the relevance of direct genetic
and maternal environmental influences to sustained genetic changes in BW
and positively correlated traits in farmed white-tailed deer populations.

A Viral-Human Interactome Based on Structural Motif-

Abstract

Protein interactions between a pathogen and its host are


fundamental in the establishment of the pathogen and underline the
infection mechanism. In the present work, we developed a single predictive
model for building a host-viral interactome based on the identification of
structural descriptors from motif-domain interactions of protein complexes
deposited in the Protein Data Bank (PDB). The structural descriptors were
used for searching, in a database of protein sequences of human and five
clinically important viruses; therefore, viral and human proteins sharing a
descriptor were predicted as interacting proteins. The analysis of the host-
viral interactome allowed to identify a set of new interactions that further
explain molecular mechanism associated with viral infections and showed
that it was able to capture human proteins already associated to viral
infections (human infectome) and non-infectious diseases (human
diseasome). The analysis of human proteins targeted by viral proteins in
the context of a human interactome showed that their neighbors are
enriched in proteins reported with differential expression under infection
and disease conditions. It is expected that the findings of this work will
contribute to the development of systems biology for infectious diseases,
and help guide the rational identification and prioritization of novel drug
targets.

Abstract

The posttreatment entomological surveillance (ES) of


onchocerciasis in Latin America requires quite large numbers of flies to be
examined for parasite infection to prove that the control strategies have
worked and that the infection is on the path of elimination. Here, we report
a high-throughput automated DNA isolation of Onchocerca volvulus for PCR
using a major Latin American black fly vector of onchocerciasis. The
sensitivity and relative effectiveness of silica-coated paramagnetic beads
was evaluated in comparison with phenol chloroform (PC) method which is
known as the gold standard of DNA extraction for ES in Latin America. The
automated method was optimized in the laboratory and validated in the field
to detect parasite DNA in Simulium ochraceum sensu lato flies in comparison
with PC. The optimization of the automated method showed that it is
sensitive to detect O. volvulus with a pool size of 100 flies as compared with
PC which utilizes 50 flies pool size. The validation of the automated method
in comparison with PC in an endemic community showed that 5/67 and
3/134 heads pools were positive for the two methods, respectively. There
was no statistical variation (P <0.05) in the estimation of transmission
indices generated by automated method when compared with PC method.
The fact that the automated method is sensitive to pool size up to 100
confers advantage over PC method and can, therefore, be employed in large-
scale ES of onchocerciasis transmission in endemic areas of Latin America.

Alkaline pretreatment of Mexican pine residues for


bioethanol production

Abstract

The locally sourced residue samples of Pinus arizonica, Pinus


cooperi, and Pinus durangensis from the state of Durango in Mexico were
analyzed for optimal yield of ethanol production. The samples were mixed
at an equal proportion using a particle size of 0.59 mm. Each individual
mixture was pretreated with either NaOH or Ca (OH)2 (at 0.5, 1.0 and
1.5% w/v) for periods of 30, 60, and 90 min at 60, 90, and 120C. The
pretreated blending was subjected to enzymatic hydrolysis for 130 h at 80
rpm and 50C with an enzymatic load of 25 filter paper units (FPU) and
50 IU -glucosidase per gramme of cellulose to obtain a maximum yield of
reducing sugars (RS) with NaOH subject at 120C for 90 min. The results
show that the hydrolysis yield depends on temperature and alkali
concentration particularly (NaOH), which increased from 2.0 to 3.5% w/v.
The best yield of glucose (41.33% w/w) was obtained using a pretreatment
of 2.5% NaOH for 90 min, 120C, and a hydrolysis residence time of 130
h. The removal of lignin and hemicellulose acetylation was observed to
have influence on the enzymatic digestibility of cellulose. This process
could theoretically produce a maximum yield of 90.19% of ethanol /
substrate (glucose) and about 80 L of bioethanol per dry ton of woody
biomass from pine residues.

Abstract

The channel catfish farming in Mexico began in the 1970s. One


of the most important areas of production is its northeastern region.
Traditionally, the channel catfish farmers have attempted to preserve their
original genetic stock. After more than 30 years of production, the genetic
relationship between stocks has not been assessed. The goal of this study
was to evaluate the genetic differentiation between five channel catfish farm
populations, using 13 microsatellite markers. A total of 154 fin clip samples
were collected from five channel catfish farms in Tamaulipas. We also
included 31 individuals of the commercial strain NWAC103. In each
population, we calculated parameter of genetic variability: inbreeding
coefficient (FIS), pairwise differentiation (FST) and genetic distance (DC). For
the inference of populations and individual assignment two Bayesian
methods were used. Genetic variability parameters were NA= 9.54 to 11.08
and AE=5.40 to 6.67. Hardy-Weinberg equilibrium deviations were observed
for all farm populations examined, and a deficit of heterozygotes was found.
Signals of inbreeding were observed, particularly for the ACU hatchery.
Populations EKA and ACU showed higher genetic differentiation values with
respect to the commercial strain (FST=0.107; FST=0.082, respectively).
Moderated genetic differences (FST=0.067; DC=0.080) were observed
between the EKA and ACU. Clustering analysis revealed a well-defined group
between EKA and TPA. In the Bayesian analysis three groups were defined.
The first group was formed by EKA and TPA, the second group included
ACU, LAJ and PRO. In the third group was the commercial strain. In the
assignment of individuals, EKA and ACU were represented by exclusive
genotypes. Identification of two genetic stocks, one located in the EKA and
the other in the ACU population, will be relevant for future management of
these hatcheries. Additionally, Page 2 of 36 Journal of the World

presented will be important tools for genetic monitoring of these farm
populations.

Abstract

In our search for new antiamoebic agents, a new series of ethyl


and methyl quinoxaline-7-carboxylate 1,4-di-N-oxide derivatives have been
synthesized using the Beirut reaction. All compounds were characterized by
spectroscopic techniques and elemental analysis. Antiamoebic activity was
evaluated in vitro against Entamoeba histolytica strain HM1:IMSS by the
microdilution method, and the structureactivity relationship was analyzed.
We found that eleven quinoxaline derivatives showed greater activity than
metronidazole and nitazoxanide with IC50 values in the range 1.990.35
lM. Compounds T-001 and T-016 shows IC50 values of 1.41 and 1.47 lM,
respectively, with a value of selectivity index >60.

Abstract

The chitinase gene was molecularly characterized in five


Bacillus thuringiensis Mexican isolates,MR10,MR11, MR21, MR33, and
RN52. The proteins derived from these genes were tested for their chitinase
activity using fluorogenic chitin derivatives. In order to verify if chitinase
genes were functional, they were cloned, and enzymatic activity of
recombinant chitinases was also tested. Results indicated that enzymes
exhibited endochitinase activity. The highest hydrolytic activity shown
against the chitin tetrameric derivative occurred at pH value of 6.5, and the
optimum activity temperature was around 60 C. The recombinant
endochitinases showed a molecular mass of 77 kDa with isoelectric points
from 6.5 to 7.0. Analysis of the nucleotide sequences showed highly
conserved sequences among all isolates (9799 %). Gene sequence analysis
revealed a putative promoter (35 TTGAGA and 10 TTAATA) and a Shine
Dalgarno sequence (5-AGGAGA-3) upstream from the open reading frame.
The deduced amino acid sequence revealed that the proteins are modular
enzymes composed by a family 18 glycosyl hydrolase domain located
between amino acids 134 and 549, a fibronectin-binding domain (580
through 656), and a chitin-binding domain (664 through 771). The deduced
amino acid sequences of our isolates showed a similarity close to 100 %
respect to the sequences reported in the GenBank database.

Visual and Gustatory Responses of Spodoptera Frugiperda

Abstract

Visual and gustatory responses of Spodoptera frugiperda


Hbner (Lepidoptera: (Noctuidae) larvae to artificial food dyes were
studied. The objective was to etermine the ability of these substances to
attract and elicit feeding of insect larvae to assess their feasibility for
improving the efficacy of ingested insecticides. The artificial food dyes
lemon green, lemon yellow, orange red, chlorophyll and carmine red were
used. Different blends were prepared with each dye to evaluate larval
visual responses to color, larval preferences to color compared with a
control, and larval gustatory responses. Only lemon green and lemon
yellow dyes visually evoked the attraction of significant percentages of the
larvae, but the other colorants did not elicit responses different from the
control. Gustatory response tests indicated that larvae preferred to feed
on gelatinized blends containing chlorophyll. The gelatinized blend
containing lemon green dye was a strong gustatory repellent, and none of
the other dyes evoked negative gustatory responses. Adding the artificial
food dye lemon green to an insecticidal formulation could enhance insect
attraction, while adding chlorophyll could enhance ingestion of the
formulation. The study of sensory aspects of insect behavior can be useful
for maximizing the efficiency of insecticidal formulations.

El da 4 de julio se llev a cabo la Ceremonia de Graduacin


Generacin 2011 2013, de la Maestra en Ciencias en
Biotecnologa Genmica, donde se les entreg las constancia de

Con motivo de la Independencia de Mxico, la Direccin de este


Centro organiz una comida mexicana para celebrar este festejo el
da 13 de septiembre, as mismo la Dra. Ninfa Rosas dio la
Bienvenida a los alumnos de nuevo ingreso a la Maestra en
Ciencias en Biotecnologa Genmica, presentando a cada
estudiante indicando el lugar de procedencia de los mismos.

Cumpliendo 15 aos
El da 15 de agosto se hizo entrega del reconocimiento por Estmulo
de Antigedad que otorga la SEP al personal de Apoyo y Asistencia a
la Educacin por cumplimiento de 15 aos de servicio efectivo a la C.
Hilda Seplveda Gonzles y C. Aleyda Flores Solis. Felicidades!
